摘要
将前期体外筛选获得的具有良好抗氧化功能的鼠李糖乳杆菌LVg、嗜热链球菌grx02、屎肠球菌ST1、婴儿双歧杆菌BF以及普通菌株鼠李糖乳杆菌C8M(普通对照)进行急性酒精性肝损伤小鼠试验,各乳酸菌组按每千克体重胃管灌服含乳酸菌数为108、107、106cfu.mL-1的脱脂乳发酵液10 mL,连续15 d,第15天时与模型组同时按每千克体重灌服50%酒精12 mL,对照组灌等量蒸馏水,禁食12 h后处死,检测血清中谷氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、白蛋白(ALB)、总蛋白(TP)及肝匀浆中丙二醛(MDA)、还原性谷胱甘肽(GSH)、超氧化物歧化酶(SOD)、乙醇脱氢酶(ADH)水平,观察肝脏病理学变化。结果表明:与模型组比较,乳酸菌组ALT、AST、MDA水平显著降低,ALB、TP、GSH、SOD、ADH水平显著升高。4株优选乳酸菌保肝效果明显优于对照组,且grx02组总体效果最佳,某些指标(MDA、SOD)上保护效果虽与空白组无显著差异,但优于药物组。
The hepatoprotective effects of LAB (Lactobacillus rhamnosus LVg, Streptococcis thermophilus grx02, Enterococcus faecium ST1, Bifidobacterium lactis BF, L. rhamnosus C8M ) were evaluated in vivo using acute ethanol- intoxicated rats as an experimental model. LAB fermented skim milk (10^8, 10^7, 10^6 cfu . mL^-1) were given at a dose of 10 mL.kg^-1 bw per mouse through gastric gavaging for 15 days, the control group was given water. The mice models of acute hepatic injury were also established by 50% alcohol (12 mL.kg%-1) after 15 d. The control group only used distilled water. The mice were fasted for 12 h, then killed. Serum levels of AST, ALT etc were measured; SOD and MDA etc in liver tissue as well as hepatic pathological changes were also observed. LAB groups decreased obviously the levels of serum AST, ALT (P〈0.05) and deceased the content of MDA, and increased the levels of SOD, GSH and ADH (P〈0. 05, P〈0. 01) in liver tissue. The protective effects of LAB were also proved by liver pathology changes. The combined effects of S. thermopilus grx02 offered protection against alcohol-induced oxidative stress, which was better than other screened LAB, superior to drug group and close to control group, while the beneficial effects of the screened LAB were obvious compared with LAB control group C8M.
