原发性肝癌患者免疫原快速激活红细胞调控IL-8和球蛋白变化仿真实验研究 被引量:2

Immunogen-activated erythrocytes modulate the release of IL-8 and immuneglobulin (Ig) in the plasma of primary hepatocarcinoma

摘要 目的研究厅发性肝癌患者血浆中红细胞固有免疫反应与来自白细胞的IL-8和球蛋白之间规律变化的关系。方法灭活癌细胞(S180株5×10^6/m1)或生理盐水(对照组)0.2ml加到枸橼酸抗凝的0.2ml全血细胞悬液(自然实验组)或白细胞悬液(白细胞分离实验组)和新鲜血浆0.3ml中,混匀后放37℃温育1h,测定反应液中IL-8和球蛋白含量。结果原发性肝癌患者自然实验组IL-8含量(376.35±243.96)pg/ml和球蛋白(7.86±2.01)g/L,明显高于正常人自然实验组的IL-8含量(11.36±6.93)pg/ml和球蛋白含量(7.21±1.75)g/L,(P〈0.01),正常人自然实验组IL-8含量(11.36±6.93)pg/ml和球蛋白含量(7.21±1.75)g/L明显高于正常人自然对照组IL-8含量(4.98±4.35)pg/ml和球蛋白含量(5.38±1.61)g/L,(P〈0.05),正常人自然实验组IL-8的激活指数2.49±2.33明显高于原发性肝癌患者自然实验组IL-8的激活指数0.22±0.24,(P〈0.05)。结论红细胞能快速调控血浆中的IL-8和球蛋白含量,但原发性肝癌患者红细胞调控IL-8和球蛋白的能力明显低于正常人,在原发性肝癌患者系统血液固有免疫反应中,红细胞固有免疫反应活性与白细胞固有免疫反应活性之间的关系处于失衡状态。 Objective To study the relationship between erythrocyte innate immune response and the release of IL-8 and Ig trom white blood cells in the plasma of patients with primary hepatocarcinoma (PHC). Methods Cancer ctlls (S180 5 × 10^6/ml) or saline ( contort group) were added to anticoagulant suspension of whole. The n ixture 0.2 ml blood cells( nature experimental group) or white blood cells (white blood cell isolation experimental group) treated with citric acid and 0.3 ml plasma, was incubated at 37℃ for l h. The level of IL-8 and immuneglobulin (Ig) in the reaction was detected. Results The leve of IL-8 (376.35 ± 243.96) and the level of Ig (7.86 ± 2.01 ) in nature experimental group of PHA were significantly higher than IL-8 ( 11.36 ± 6.93 ) and Ig (7.21± 1.75 ) in nature experimental group of normal control ( P 〈 0. 01 ). The level of IL-8 ( 11.36± 6.93 ) and Ig (7.21±1.75 ) in nature experimental group of normal control was s gnificantly higher than IL-8 (4.98 ±4.35) aM Ig (5.38 ±1.61 ) in nature control group of normal contvl (P 〈0.01 ). Activation index(AI) of IL-8 (2.49 ± 2.33 ) in nature experimental group of narmal control was significantly higher than Al of IL-8 (0.22±0.24 ) in nature experimental group with PHA (P 〈0.05 ). Conclusion In systemic blood quick innate immunoreaction, the erythrocytes can quickly regulate the level of IL-8 and Ig in plasma. The capatity of erythrocytes to regulate IL-8 and Ig in patients with PHC was much lower than that in normal people. In patient with PHC, the relationship between red blood cell innate immunity and white blood cell innate immunity in plasma are being unbalanced.
