摘要
为建立东方百合元帅、提拔、西伯利亚的高频愈伤体系,以幼叶、花丝、生长点为外植体,进行了不同激素组合的MS固体培养基对胚性愈伤组织诱导及植株再生研究。结果发现,Picloram比2,4-D更有利于胚性愈伤组织的诱导,幼叶、花丝更能有效诱导出胚性愈伤组织和再生植株,生长点的出愈率较低。幼叶在MS+2 mg/L Picloram培养基,花丝在MS+2 mg/L Picloram+0.1 mg/L ABA培养基,生长点在MS+2 mg/L Picloram+0.03 mg/L TDZ培养基上能高效诱导出愈伤组织,来自幼叶的胚性愈伤组织在MS+1.0 mg/L BA+0.1 mg/L KT+0.1 mg/L IBA培养基上能高频再生出芽,来自花丝的胚性愈伤组织在MS+1.5 mg/L BA+0.3 mg/L KT+0.2 mg/L IBA培养基上能高频再生出芽。愈伤组织生长呈直线生长趋势。
The leaf, style and shoot tip of oriental lily were used as the exp)ants to study the embryonic callus inducement and plantlet regeneration on the solid MS medium containing different hormone combinations for establishing high frequency callus of Acapulco, Tiber and Siberia. It was found that MS medium containing Picloram was more favorable for embryonic callus formation than the MS medium containing 2,4 - D, and the leaf and style could be induced to form embryonic callus and regenerated plantlets. The leaf cultivated on tile medium of MS + 2 mg/L Picloram + 0. 1 mg/L ABA and the style cultivated on the medium of MS + 2 mg/L Pictoram + 0.03 mg/L TDZ and the shoot tip cultivated on the medium of MS + 2 mg/L Picloram could be induced to form calluses highly effectively. The calluses from the leaf could be induced on the medium of MS + 1.0 mg/L BA + O. 1 mg/L KT + 0. 1 mg/L IBA to form re- generation buds at a high frequency and the calluses from style could be induced on the medium of MS + 1.5 mg/L BA + 0.3 mg/L KT + 0.2 mg/L IBA to form regeneration buds at a relatively high frequency. The callus growth was in the trend of linear curve.
