摘要
对欧拉型藏绵羊生长激素释放激素受体(GHRHR)基因部分片段进行PCR扩增和克隆测序(GenBank Accession No:EU289218),利用BioEdit7.0.9.0、DnaSP 4.10.9和MEGA4.0等生物信息学软件对该部分序列与GenBank中普通牛、瘤牛、水牛相应序列进行比对分析,并对58只欧拉型藏绵羊该基因片段进行SmaI-RFLP研究。结果表明:欧拉型藏绵羊与普通牛、瘤牛、水牛3个物种基因序列间同源性大小依次为92.5%、92.0%、91.5%;4个物种间共发现41处序列间碱基差异(9.65/100 bp),其中藏绵羊与普通牛、瘤牛、水牛3个牛亚科物种均存在差异碱基29处。碱基变异类型主要表现为碱基转换和颠换,少数为碱基插入和缺失。推导的部分氨基酸序列间同源性大小依次为88.8%、88.8%、94.4%。58只欧拉型藏绵羊该基因片段SmaI-RFLP分析均为单态,表现为AA基因型。
The partial sequence of Oura-type Tibetan sheep GHRHR gene (GenBank Accession No: EU289218 ) was analyzed and compared with that of cattle (B. taurus) , zebu (B. ind/cus) and buffalo (B. bubalis) from GenBank by BioEdit7.0. 9.0, DnaSP 4. 10. 9 and MEGA 4. 0 softs. In addition, the polymorphism of partial GHRHR gene of 58 Oura-type Tibetan sheep ( Ovis aries) was analyzed by SmalRFLP method. The results showed that the homology of nucleotide sequence of the partial GHRttR gene among the Tibetan sheep and the cattle, zabu, buffalo was 92.5%, 92. 0% and 91.5%, respectively. There were 41 bases difference in 4 species (9. 65/100 bp) and 29 bases difference in Oura-type Tibetan sheep and cattle, zebu and buffalo in 425 bp segment. The main base changes were base transition and base transversion, also with some base invertions and deletions. The homology of amino acids deduced from the partial exon6 of GHRHR gene among the Tibetan sheep and the cattle, zabu, buffalo was 88.8%, 88. 8% and 94. 4% , respectively. The polymorphism of partial GHRHR gene of 58 Oura-type Tibetan sheep showed that there was no polymorphism in this segment, all genotypes of Oura-type Tibetan sheep studied were AA genotype.
