摘要
用采自江苏省吴江苗圃收集的6个滨梅种源叶片样品(分别标记为7、9、10、11、12和13号),分析了ISSR、RAPD2种分子标记,以探讨各种源间的遗传变异及其亲缘关系。结果表明,ISSR标记的19个多态性较好的引物共产生了137条带,其中多态性条带91条,占总条带的66.42%。条带大小300~3000bp;RAPD标记的15个多态性引物共产生99条带,其中多态性条带53条,占总条带的53.5%,条带大小为300—3000bp。基于ISSR标记的6个种源间遗传距离为0.0680—0.9479;而基于RAPD标记的遗传距离为0.0412—0.6439。2种标记的聚类结果,7、9、10、11、12号可归为一类,另外13号单独归为一类,该分子标记结果与形态变异类型相一致。
Genetic variation of Prunus maritime collected from six provenances (marked as No. 7, No. 9, No. 10, No. 11, No. 12 and No. 13) was analyzed by the methods of ISSR and RAPD. The results showed that 19 ISSR primers produced a total of 137 bands with the size of 300 - 3 000 bp, of which polymorphism bands were 91 and accounted about 66.42% of the total. Meanwhile, 15 RAPD primers generated a total of 99 bands with the size of 300 - 3 000 bp, 53 of which were polymorphism and about 53.5% of the total. Genetic distance of Prunus maritima collected from six provenances was 0. 068 0 -0. 947 9 and 0. 041 2 -0. 643 9 based on ISSR and RAPD, respectively. The cluster analysis indicated that the provenance No. 7, No. 9, No. 10, No. 11 and No. 12 were classified as one category, and the provenance No. 13 as another separate category. The category of Prunus maritime by molecular markers was consistent with the morphological variation analysis.
