天癸更年软胶囊对雌性老年大鼠脑NGF、BDNF、Trk A和Trk B表达的影响 被引量:4

Effects of Tiangui Gengnian Soft Capsule on Expression of NGF,BDNF,Trk A and Trk B in the Brain of Aged Female Rats

摘要 目的:探讨天癸更年软胶囊(含沙棘脂肪酸)对老年大鼠的神经保护和抗衰老作用。方法:选用16月龄雌性大鼠,灌胃给予不同剂量的天癸更年软胶囊180 d,同时设立雌激素对照组、老年(22月龄)对照组和青年(3月龄)对照组。应用免疫组织化学和Western blot技术,观察神经生长因子(NGF)和脑源性神经营养因子(BDNF)及其高亲和力酪氨酸激酶受体TrK A和TrK B在天癸更年软胶囊给药组和对照组大鼠脑内的表达。结果:免疫组织化学研究显示,天癸更年软胶囊可显著增加雌性老年大鼠脑白质内NGF阳性星形胶质细胞的相对面积,显著增加某些脑区NGF、Trk A、BDNF和Trk B阳性神经元数目。Western blot研究显示,天癸更年软胶囊可显著增加雌性老年大鼠脑内NGF和BDNF的相对表达量。结论:天癸更年软胶囊对老年期脑有一定的神经营养和保护作用。 Objective:To investigate the neuroprotective and anti-aging effects of Tiangui Gengnian Soft Capsule (TGSC), which is composed of seabuckthorn fatty acids, on the aged rats. Methods : Sixteen-months-old female rats were used in this study and intragastric administration of different doses of TGSC were given for 180 days. The rats at the age of 3 and 22 months were taken as controls. Immunohistochemical method and Western blot technique were carried out to observe the expression of the nerve growth factor (NGF) and brain derived neurotrophic factor (BDNF) and their respective high affinity receptor Trk A and Trk B in the brain of the TGSC-fed and control rats. Results : hnmunohistochemistry showed that TGSC significantly increased the relative area of NGF-positive astrocytes in the white matter,and the number of NGF,BDNF,Trk A- and Trk B-positive neurons in some brain regions of the aged rats. Western blot showed that TGSC significantly increased the relative expression level of NGF and BDNF in the brain of the aged rats. Conclusion :TGSC upregulates the expression of neurotrophic factors and their receptors in the brain of the aged female rats, suggesting that TGSC may have certain neurotrophic and neuroprotective effects on the brain during the aging life.
