摘要
建立了用超声波法提取蛹草拟青霉中虫草菌素和用HPLC法测定其含量的方法。试验结果表明,在检测波长259nm处,虫草菌素浓度为5.42~173.3μg/mL时,回归方程C=-1.2846+3.41192×10^-5A,r=0.9996;平均回收率100.15%,RSD=0.54%(n=4);超声波法提取菌丝体中虫草菌素时,提取溶剂以水或低于20%浓度的乙醇溶液较好,提取时间30min;用此法可测定不同蛹虫草菌株中虫草菌素的产量。
The methods of extracting eordycepin in Paecilomyces militaris and detecting cordycepin content were set up. The results showed that the method for extracting cordyeepin in Paecilomyces militaris was the ultrasonic wave under the condition of water or 〈20 % ethanol of extraction solvent and 30 min of extraction time. The eordycepin eontent was detected by HPLC under 259 nm wave-length and the regression equation C=-1.2846 + 3.41192×10^-5A was established, when the cordyeepin content was 5.42 to 173.3 μg/mL,the value of r,average recovery rate and RSD was 0.9996,100.15 % and 0.54 % (n =4) respectively.
