摘要
目的研究白细胞源性精氨酸氨基肽酶(Leukocyte-derived Arginine Aminopeptidase,L-Rap)在高糖培养的人视网膜血管内皮细胞的表达,分析L-Rap蛋白的功能。方法体外培养人视网膜血管内皮细胞,实时定量PCR检测L-Rap在高糖培养的人视网膜血管内皮细胞的表达变化,建立L-Rap的三维蛋白结构图,分析L-Rap蛋白的功能。结果L-Rap的mRNA在高糖培养的人视网膜血管内皮细胞下降,L-Rap蛋白可能对血管生成因子、免疫炎症因子以及肾素-血管紧张素系统发挥作用。结论L-Rap与糖尿病视网膜病变密切相关,可以从血管生成因子、免疫炎症因子以及肾素-血管紧张素系统这三个途径探索L-Rap对糖尿病视网膜病变的作用。
Objective To study the expression of L - Rap in human retinal vascular endothelial cells that were cultured in high glucose, and analyze the function of L - Rap protein of human retinal vascular endothelial cells. Methods Culture human retinal capillary endothelial cells with high glucose, mRNA was detected by real time PCR, 3D protein structure model of L - Rap was established, and the function of L - Rap was analyzed. Results The mRNA level of L - Rap in human retinal capillary endothelial cells that were cultured in high glucose was down - regulated. L - Rap protein may play a role in angiogenesis, immune and inflammatory factors as well as in renin - angiotensin system. Conclusion L - Rap is closely related to the diabetic retinopathy. We can explore the role of L - Rap on the diabetic retinopathy from three ways: angiogenic factors, immune and inflammatory cytokines, renin - angiotensin system.
