摘要
目的探讨急性和重复性心理应激对大鼠血清皮质酮和免疫组织中糖皮质激素受体(GR)的影响。方法60只SD大鼠分为6组,3组分别接受为期1、14和28 d束缚应激(6 h/d),相应的3个对照组大鼠不接受任何应激处理。分别测定大鼠在应激和非应激状态下血清皮质酮水平以及胸腺组织和脾淋巴细胞中GR的水平。结果3个对照组大鼠的血清皮质酮水平分别为(103.4±6.4)、(100.0±7.8)和(99.2±11.2)ng/L血清,应激状态下则分别为(624.6±52.0)、(663.1±48.1)和(673.6±26.9)ng/L血清。14和28 d应激组大鼠的胸腺GR数目均明显低于相应的对照组动物[(38.36±2.71)vs(24.48±2.00)pmol/g蛋白,(39.11±4.13)vs(23.66±2.63)pmol/g蛋白];1、14和28 d应激组大鼠脾淋巴细胞中的GR数目亦明显低于相应的对照组动物[(5 189.88±180.11)vs(4 816.88±197.72)结合位点/细胞,(5 822.38±179.27)vs(2 990.13±120.67)结合位点/细胞,(5 887.38±108.83)vs(1 820.50±161.11)结合位点/细胞]。结论急性和重复性心理应激可以增强糖皮质激素(GC)的分泌,同时下调免疫组织中的GR水平,对动物机体造成病理性应激损伤。
Objective To explore the effect of acute and repeated psychological stress on serum corticosterone and glucocorticoid receptor in rats. Methods Six groups of male SD rats were subjected to either a single day or 14, 28 days of restraint (6 hours/day) or transferred to the testing room (unstressed). In order to examine the proportions of receptors occupied and activa- ted by stress levels of cortieosterone, we measured available type Ⅱ (glucocorticoid) adrenal steroid receptor levels in immune tissues of acutely stressed and repeated stressed rats. Results The levels of serum eorticosterone in three control groups were ( 103.4±6. 4), ( 100.0±7.8) and (99. 2±11.2) ng/L serum respectively. Then the stress induced levels of serum corticoster-one in three treated groups elevated to (624. 6±52.0), (663.1±48.1) and (673.6±26.9 ) ng/L serum respectively. The numbers of glucocorticoid adrenal steroid receptors of thymus gland in 14 and 28 d-stressed groups were lower than those of the control groups [(38.36±2.71)vs(24.48±2.00) pmol/g protein and(39. 11±4. 13)vs(23.66±2. 63)pmol/g protein]. The numbers of of spleen lymphocyte receptors in three stressed groups were also lower than those of the control [ (5 189.88±180. 11 )vs(4 816. 88±197.72) sites/cell, ( 5 822. 38±179. 27 ) vs (2 990.13±120.67) sites/cell and (5 887. 38±108.83 )vs(1 820. 50±161.11 )sites/cell]. Conclusion Acute and repeated psychological stress are associated with the stress-induced impairment through strengthening the secretion of glucocorticoid and downregulation the level of glueocorticoid adrenal steroid receptor.
