摘要
对滇藏地区8种鹅膏菌的核糖体DNA(rDNA)内转录间隔区(ITS)进行PCR扩增和测序,用Clustal X(version1.81)软件对所测的ITS序列进行比对分析.结果表明,除5.8SrDNA(其长度为160bp)比较保守外,8个不同种的ITS序列在长度和碱基位点上都存在较大差异,其中ITSI区间的片段长度为206~297bp,ITS2区间的片段长度为191—238bp.这说明鹅膏属的种间具有较高的遗传多样性.利用MEGA(version3.1)软件进行聚类,8个鹅膏菌在D=0.14处分成3组,与传统分类有出入.这为鹅膏菌的进一步分类及研究提供了分子依据.
The internal transcribed spacer (ITS) regions of ribosomal DNA of 8 Arnanita species from Tibetan and Yunnan in China were sequenced by PCR amplication, and their sequences were analyzed by the software of Clustal X( version 1.81 ). The results showed that, the ITS sequences of 8 species had significant differences in both length and bases, besides the fragment of 5.8S rDNA , whose length is 160 bp, being relatively conservative; the length of these fragments of ITS1, ITS2,were 206-297 bp, 191-238 bp,respectively. These implied that the genetic diversity among the Arnanita species was relatively high. The 8 Amanita species was dividied into 3 groups at D = 0.14 according to the phylogenitic analysis by MEGA and it was dissimilar to traditional taxonomy. The results above provided a molecular reference for further taxonomy and study of Amanita in the future.
