摘要
介绍免疫磁珠分离技术的原理和操作步骤,分析其主要影响因素。介绍该技术在环境样品病原菌、原虫及病毒检测中的应用及研究进展。环境样品成分复杂,需经适当分离纯化处理再进行病原微生物检测,从而保证检测结果的可信性。免疫磁珠分离技术兼具免疫反应的特异性及磁性分离的快速性,能够有效分离纯化环境水样中的病原微生物,广泛应用于病原微生物的检测。
This paper is aimed to present the main procedure of Immunomagnetic Separation (IMS) and discuss its main primary influential factors. As is well known, IMS is an emerging technique for its virtue of specificity and celerity, which has been successfully adopted to isolate pathogens from samples. In addition, IMS greatly relies on the specific antibodies coated on the surface of magnetic beads to capture the targeted microorganisms, and subsequently separate them from samples in the presence of a magnetic field. As a matter of fact, IMS has already found successful application in the detection of pathogenic bacteria, since it performs well in removing inhibitors and non-targeted microorganisms that influence the examination, thus sat- isfying the demands of specific separation and high efficient purification of the targeted bacteria in samples. Moreover, IMS can be easily combined with other immunological and molecular biological methods in detecting pathogenic bacteria, which greatly improves the detection sensibility and has greatly shortened the detection time. Besides, it is also widely employed in the detection of parasites, Much as Giardia and Cryptosporidium, as an agent to speed up the separation of the oocysts/cysts from the concentrated debris. Therefore, compared with such separation methods as flotation, IMS has been found to be superior, for it demonstrates better separation efficiency and thus can reduce the incidence of false-positives. In addition, it has also been introduced for detection of viruses. Especially, the combination of IMS and other techniques such as PCR may allow for the detection of antigen and nucleic acid and thereby is expected to provide a better correlation with viral infectivity than either method alone. All in all, its application and the latest researches on it in the detection of pathogenic bacteria, parasites and viruses would surely be paid more and more attention from the science circle.
