两种新型倍半萜类化合物诱导人肝癌细胞SMMC-7721凋亡(英文) 被引量:2

摘要 研究两种从盘花垂头菊中分离出的高含氧甜没药烯型倍半萜(HOBS)化合物对肿瘤细胞的影响.结果显示HOBS能够诱导人肝癌细胞SMMC-7721凋亡.单细胞电泳实验证明HOBS能够以剂量依赖的方式诱导SMMC-7721细胞损伤,流式细胞技术显示HOBS能够以剂量依赖的方式诱导SMMC-7721细胞凋亡,凋亡率达到38.9%,c_i(Ca^(2+))的测定显示HOBS能够诱导c_i(Ca^(2+))升高,所有的结果证明HOBS能够诱导人肝癌细胞SMMC-7721凋亡.提示这两种化合物有抗人肝癌细胞的SMMC-7721活性,为抗肿瘤药物的设计提供了新的思路. The effects of two novel highly oxygenated bisabolane-type sesquiterpenes (HOBS) isolated from C. Discoideura (Cremanthodiurn discoidiem) on tumor cells were investigated. The results indicate that HOBS induced DNA damage and apoptosis of SMMC-7721 cells both in a dose-dependent manner as revealed by single cell gel electrophoresis and flow cytometry analyses, and the apoptosis rate was up to 38.9%. Measurement of ci(Ca^2+) showed that HOBS could induce elevation of Ca^2+. All the results suggest that HOBS could induce SMMC-7721 cells apoptosis.
