摘要
目的研究他克莫司(TM)对长波紫外线(UVA)照射后HaCaT细胞表达细胞间黏附分子-1(ICAM-1)的影响,以探讨TM治疗光敏性皮肤病的作用机制。方法采用ELASI和免疫组化法检测在不同强度UVA照射后,HaCaT细胞表达ICAM-1的情况。结果与未经UVA照射组细胞比较,未加TM组经UVA照射24h后,HaCaT细胞表达ICAM-1水平明显升高(P<0.01),而加入TM组的ICAM-1表达明显受到抑制(P<0.01)。结论TM对UVA照射后HaCaT细胞表达ICAM-1有抑制作用,提示对UVA引起的炎症有抑制作用。
Objective To investigate the effect of tacrolimus (TM)in the expression of ICAM-1 on the HaCaT cell after UVA exposure and discuss the mechanism of action on tacrolimusin photoallergic dermatitis. Methods The expression of ICAM-1 on the HaCaT cell were measured by ELASI and immunohistochemistry after UVA exposure with different intensity. Results The group in that the UVA exposure was practiced for 24h had significantly more ICAM-1 expression on HaCaT cell that in the group without UVA exposure. The use of TM showed to inhibit significantly this the espression of ICAM-1. Conclusion The use of TM had an inhibitory effect of the expression of ICAM-1 on the HaCaT cell after the UVA expresure.
