摘要
以菠萝幼果为研究材料,采用Creator^(TM) SMART^(TM) cDNA Library Construction Kit技术构建菠萝幼果cDNA文库,其文库的库容量为1.01×10~6,重组率为92%,PCR检测表明大多数插入片段均大于1 000 bp。
Total RNAs were extracted from the pineapple fruit using plant total RNA extraction kit, and fulllength cDNA library was constructed with SMART cDNA Library Construction Kit. The capacity of the primary library was 1.01×10^6, in which 92% clones were recombinant and all of the inserted cDNA fragments were larger than 1 000 bp. The positive signals were detected by PCR in the amplified library. The data indicated that the cDNA Library had high quality, and provided a useful tool for cloning glucose metabolism genes and further study on the molecular mechanisms of its biosynthesis.
