摘要
目的观察大鼠去势后体温改变与下丘脑视前区5-羟色胺2A(5-HT2A)受体亚型表达之间的关系。方法用数字温度计TM902C检测大鼠的肛温,酶联免疫吸附试验测定血清激素水平,根据5-HT2A受体互补DNA序列合成相应的特异性引物,用PCR法观察大鼠去卵巢后下丘脑视前区5-HT2A受体表达。结果①大鼠去势组(OVX)自第八周末肛温明显高于假去势(sham)组(P<0.05)。②OVX组大鼠血清17β-雌二醇(17β-estradiol,E2)较sham组下降,LH水平升高,差异显著(P<0.05)。③OVX组下丘脑视前区5-HT2A受体表达增强,与sham组比较有显著性差异(P<0.05)。结论大鼠卵巢摘除后体温升高,下丘脑视前区5-HT2A受体表达增强,提示5-HT在低雌激素状态下的体温升高调节中起一定作用。
Objective To observe the relationship between temperature change and the expression of 5 HT2A recep tor subtype in preoptic area and anterior hypothalamus of ovarieetomized(OVX) rats. Methods The digital thermometer TM902C was used to measure rectal temperature (RT). Enzyme-linked immunoadsordent assay (ELISA) was used to detect serum hormone levels. The method of polymerase chain reaction (PCR) was used to detect the presence of 5-HT2Areceptor subtype in the preoptic area and anterior hypothalamus of rat after ovariectomy. The specific olig-onucleotide primers were synthesized based on the complementary DNA sequences for the rat 5-HT2Areceptor subtype. Results ①The RT in OVX rats was significantly elevated 8 weeks after ovariectomy compared with those in sharn operated control group (P〈0. 05). ②Compared with the control group, concentrations of serum E2 decreased in the OVX group (P〈0. 05), while concentration of LH inereased (P〈0. 05). ③The expression of 5-HT2A receptor subtype in preoptic area and anterior hypothalamus of the OVX rats was significantly higher than control group. Conclusion The RT was elevated after ovariectomy and the expression of 5-HT2A receptor subtype in preoptic area and anterior hypothalamus of the OVX rats were significantly higher. Serotonin may play some roles in thermoregulation in the state of low hormone eoneentrations.
