摘要
目的建立血清中茶碱的HPLC测定方法,开展茶碱血药浓度临床监测。方法采用15%的高氯酸处理血样,色谱柱为Shim—pack VP—ODS(150×4.6mm,5um),流动相为甲醇:水=28:72,检测波长为274nm,流速1mL/min,内标为咖啡因。结果茶碱标准曲线回归方程为Y=1.12742X-0.0173523,r=0.9997(n=5),线性范围为1—40ug/mL。茶碱5、15、30ug/mL提取回收率分别为75.1%、73.9%、74.5%,方法回收率为102.3%、99.0%、99.8%,日内RSD(相对标准差)分别为5.8%、2.8%、3.5%,日间RSD分别为4.1%、3.4%、2.6%。结论本测定方法简便、快速、准确,适合茶碱临床血药浓度监测。
Objective To develop a method for the determination of theophylline concentration in human serum by HPLC and carry out therapeutic drug monitoring (TDM) of theophylline in clinical. Methods The serum sample was extracted with 15% perchloric acid, then separation was achieved on the Shim - pack VP - ODS column ( 150 × 4.6mm, 5um). The mobile phase was consisted of methanol - water = 28:72, the effluent was measured at 274nm and the flow rate was 1 mL/min. Caffeine was used as the internal standard. Results The regression equation of theophylline was Y = 1. 12742X - 0. 0173523, r = 0. 999 7 ( n = 5). The calibration curves of theophylline were linear over the range of 1 -40 ug/mL. The extraction recovery of 5, 15, 30 ug/mL theophylline was 75.1%, 73.9 % and 74.5 % respectively. The method recovery was 102.3%, 99.0% and 99.8%. The inter-day RSD was 5.8%, 2.8% and 3.5%. The within - day RSD was 4.1%, 3.4% and 2.6%. Conclusion This assay was simple, rapid, and accurate way to measure the concentration of theophylline in human serum. It is suitable to be used for TDM of theophylline in clinical.
