摘要 目的:观察糖尿病大鼠下颌下腺内凋亡相关基因Fas和FasL的表达变化。方法:SD雄性大鼠随机分为4周、12周模型组及对照组。用链脲佐菌素复制出糖尿病动物模型,分别于第4、12周后测体重和血糖,并取下颌下腺组织,应用免疫组织化学方法检测Fas及FasL表达变化,计算机图像分析系统测平均光密度值(MOD)。结果:大鼠下颌下腺内有Fas及FasL表达,分布于颗粒曲管、纹状管及小叶间导管上皮细胞胞质及部分细胞核内。对照组呈Fas及FasL弱阳性,4周模型组多呈中等阳性,部分呈强阳性;12周模型组大多呈强阳性。与对照组比较,4周、12周模型组各级导管上皮细胞内Fas及FasL MOD值均增高(P<0.01)。结论:大鼠下颌下腺内Fas及FasL表达随糖尿病病程延长而增强,可能在糖尿病时下颌下腺组织凋亡中起重要作用。 Objective To investigate the expression of apoptosis-associated genes Fas and FasL in submandibular gland of diabetic rats. Methods Male Sprague-Dawley rats were randomly divided into 4 weeks model group (n=6), 12 weeks model group (n=6) and normal control group (n=12). The diabetic rats models were induced by injection of streptozotoein into peritoneal cavity. After 4 and 12 weeks, animals were killed for assaying blood sugar and weighing body weight. The submandibular gland were observed by immunohistochemistry methods, and then the optic density of Fas and FasL immunoreactive cells was detected by computerized image pattern analysis. Results The Fas and FasL were expressed in submandibular gland of all groups, and was distributed in the cytoplasma and nuclei (partially) of the epithelial cells of granular convoluted tubule, striated and interlobular duct. The expression of Fas and FasL showed weak positive in normal control group, moderate in 4 weeks model group and intensive in in 12 weeks model group. Compared with normal control group, mean optical density level of Fas and FasL immunoreactive cells in submandibular gland increased significantly in 4 and 12 weeks model groups (P 〈 0.01 ). Conclusion The expression of Fas and FasL gradually increases in submandibular gland with diabetes progressing and may play an important role in the apoptosis of submandibular gland.
