摘要
目的:确定栀子油的最佳提取条件,观察栀子油的镇静、催眠、抗惊厥及促学习记忆作用.方法:采用超临界CO2萃取栀子油,用极差法确定其最佳工艺条件;利用光电计数法观察灌胃给予栀子油对小鼠自发活动的影响,以及长期使用栀子油对小鼠自发活动耐受性的影响;利用延长戊巴比妥钠睡眠时间实验和戊巴比妥钠阈下催眠剂量实验,观察栀子油的催眠作用;利用戊四氮实验观察栀子油的抗惊厥作用;利用避暗、跳台实验,观察栀子油的促学习记忆作用;采用地西泮作为平行对照.结果:萃取压力是影响栀子油得率的主要因素,萃取栀子油得率12.0%,其最佳工艺条件是:萃取压力25MPa;萃取温度45℃;解析釜Ⅰ压力12MPa;解析釜Ⅰ温度45℃.栀子油可抑制小鼠自主活动,此作用随用药剂量(1~10mL/kg)增加而增强,随用药时间延长未出现明显耐受现象;栀子油可诱导小鼠入睡,延长睡眠时间,与戊巴比妥钠有协同作用;延长戊四氮引起的小鼠惊厥潜伏期;减少小鼠入室错误次数.结论:超临界CO2萃取的栀子油具有镇静、催眠、抗惊厥及促学习记忆的作用.
AIM: To explore the optimal extraction condition of gardenia oil and to investigate its effect on sedation, hypnosis, anti-seizure, the ability of learning and memory in mice. METHODS: Supercritical CO2 extraction was used to extract oil from gardenia and the best qualification was decided by direct-viewing analysis. Photo-cell method was used to observe the effect of the oil on the spontaneous activity and tolerance after a long-term dose by intragastric administration in mice. Hypnosis effect of the oil was investigated under two experiments, the sleeping-time above threshold dose and sleeping number below threshold dose using pentobarbital sodium in mice. The effect of anti-seizure of the oil in mice was inspected by pentetrazole. The function of the oil on the ability of learning and memory of mice was observed by step down test and step through test. Diazepam was used as standard control in all experiments. RESULTS: Extraction pressure was the main factor affecting extraction yield which approached 12% in the experiment. The requirement of the optimal extraction technology condition was as follows : pressure 25 MPa ; temperature 45℃;analysis caldron Ⅰ pressure was 12 MPa; analysis caldron Ⅰ temperature was 45℃. Gardenia oil inhibited the spontaneous activities of mice. The sedation effect increased gradually without any tolerance with the increasing dose and time of the drug taken. It had evident hypnosis effect for being able to prolong sleepingtime, increase sleeping number and had synergy effect along with pentobarbital, h prolonged the seizure delitescence of mice by pentetrazole and decreased error number of mice in step through test. CONCLUSION:Gardenia oil by supercritical CO2 extrac-tion has the function of sedation, hypnosis and anti-seizure, as well as promoting the ability of learning and memory in mice.
