摘要
目的了解丙型肝炎病毒(HCV)非结构区蛋白质NS3与干扰素调节因子3(IRF3)的相关关系。方法①建立病毒感染条件下IRF3核内移的实验室检测方法;②构建包含HCV NS3基因片段的质粒;③建立免疫荧光检测方法,分析NS3对IRF3核内移的影响,进而明确二者的关系。结果病毒感染和HCV NS3同时作用可以明显降低IRF3的核内移率。结论HCV NS3具有调节IRF3功能的作用。HCV有可能通过抑制IRF3来实现病毒免疫逃避。
Objective To investigate the relationship between the HCV NS3 protease and interferon regulatory factor 3 (IRF-3). Methods In the present study, first set up the experiment for the detection of IRF, then the plasmid containing HCV NS3 genome was constructed. Finally the relationship between the NS3 and endogenous IRF-3 was studied. Results IFA results showed that comparing with activating effect of virus infection alone, collective action of transfection and virus infection could more effectively reduce the nuclear translocation of endogenous IRF-3 of HeLa cell. Conclusion HCV NS3 protease displayed obvious inhibition of nuclear translocation of endogenous IRF-3.
