摘要
目的观察牛磺酸对大鼠视网膜缺血再灌注损伤(retinal ischemia-reperfusion,RIR)的影响及其作用机制。方法将120只大鼠随机分为3组:对照组、缺血组、保护组,采用前房灌注液体形成14.63 kPa高眼压1 h的方法,建立RIR模型,连续3 d,2次/d,腹腔注射10%的牛磺酸,剂量为100 mg/kg,于手术前30 min加注1次。对照组给予同等剂量的生理盐水。两组缺血60 min后,分别再灌注0、2、6、12、24、48、72 h用比色法进行视网膜SOD、MDA、NO的测定;用光镜测量包埋切片的平均视网膜内层厚度(mean thickness of inner retinal layer,MTIRL)。结果牛磺酸能显著对抗大鼠视网膜缺血再灌注后MDA和NO水平的升高,同时能改善平均视网膜内层厚度的变化,但对SOD的作用不确定。结论牛磺酸可减轻大鼠视网膜缺血再灌注后的损伤。
Objective To investigate the effect of taurine on retina in rats with the damage of retinal ischemia-reperfusion and its mechanism. Methods 120 Wistar rats were divided randomly into 3 groups: control group, ischemia group, protective group. Increasing the intraocular pressure to 110 mm Hg for 60 minutes with anterior chambers infused liquid induced the models of retinal ischemia-reperfusion of rats, 3 days, 30 minutes before retinal ischemia, the rats in the protective group received a intraperitoneal injection of taurine at dose of 100 mg/kg twice a day for 3 days and the rats in the control group received a intraperitoneal injection of sodium chloride at same dose , at 30 minutes, 12 hours,2g hours,g8 hours,72 hours after ischemia- reperfusion, the effect of taurine on retinal degeneration was assessed by measuring the level of SOD, MDA, NO and the thickness of the inner retinal layers(MTIRL) . Results The level of SOD was decreased,but those of MDA and NO both were increased after ischemia-reperfusion retina of rats. the changes of MDA and NO were blocked by taurine. but the effect of tanrine to SOD was not confirmed. Conclusion Taurine can protect the retine from ischemia-reperfusion injury.
