摘要
目的探讨扩张型心肌病(DCM)患者外周血单个核细胞(PBMCs)核转录因子кappaB(NF-кB)/p65的表达及阿托伐他汀钙(Ale)的干预作用。方法对25例DCM患者和18名健康对照者分离出的PBMCs进行培养,共培养24h,并将培养的PBMCs分成3组:空白对照组、脂多糖(LPS)组和Ale组。对3组培养后的PBMCsNF-кB/p65的表达进行免疫组化染色检测和半定量分析。结果DCM患者PBMCsNF-кB/p65胞核染色阳性率明显高于健康对照者[(14.86±3.21)%比(6.32±2.25)%,P<0.01];脂多糖组DCM患者和健康对照者PBMCsNF-кB/p65胞核染色阳性率与空白对照组比较均显著增加(P<0.01);Ale组DCM患者和健康对照者PBMCsNF-кB/p65胞核染色阳性率与脂多糖组比较则显著下降(P<0.01)。结论DCM患者NF-кB/p65表达增高,而Ale可下调脂多糖刺激下NF-кB/p65表达增高。
Objective To investigate the expression of nuclear factor κB/p65 on peripheral blood mononuclear cells (PBMCs) in dilated cardiomyopathy (DCM) patients and effect of atorvastatin calcium (Ale). Methods PBMCs isolated from peripheral blood in DCM patients(n=25) and healthy subject (n=18) were divided into 3 groups: control group, lipopolysaccharide (LPS) stimulate group and Ale group, and cultured 24 hours in 1640 medium. The expression of NF-κB/P65 in PBMCs were measured by immunocytochemical staining. Results Compared with the healthy subjects, the expression of NF-κB/P65 on PBMCs in DCM patients was found significontly increased (14.86±3.21% vs 6.32±2.25% ,P〈0.01).In LPS stimulation, the expression of NF-κB/P65 on PBMCs in DCM patients and healthy subject was found significontly increased(P〈0.01 ). Ale significantly inhibited this increment in DCM patients and healthy subject (P〈0.O1). Conclusion The expression of NF-κB/P65 in DCM patients is found significantly increased, but Ale can inhibit LPS stimulating increment of NF-κB/P65.
