摘要
为了证实人脐静脉内皮细胞是否具有合成一氧化氮的能力,用NADPH-黄递酶组织化学法和原位杂交技术,对体外培养的人脐静脉内皮细胞表达一氧化氮合酶进行了观察。结果显示,体外培养人脐静脉内皮细胞的NADPH-黄递酶组织化学的阳性百分率为62%,诱生型一氧化氮合酶的原位杂交的阳性百分率为74%。阳性反应产物分布在核周及细胞突起中。本研究首次证明体外培养的人脐静脉内皮细胞能表达诱生型一氧化氮合酶,即人脐静脉内皮细胞在体外培养或受刺激情况下,具有合成一氧化氮的能力。这可能是无神经支配的人脐静脉的一种重要调节因素。
his experiment was designed to investigate whether the human umbilical vein endothelial cells have the ability to synthesize nitric oxide by means of NADPH diaphorase histochemistry and in situ hybridization. It showed that the cultured human umbilical vein endothelial cells were nitric oxide synthase positive by NADPH diaphorase histochemical staining and could express inducible nitric oxide synthase mRNA by in situ hybridization. 62% and 74% of the cultured human umbilical vein endothelial cells were positive by NADPH diaphorase histochemistry and in situ hybridization respectively, but the intensity of positive granules varied widely. This study provides the evidence that inducible nitric oxide synthase mRNA and nitric oxide synthase are localized in the cultured human umbilical vein endothelial cells, and shows that the human umbilical vein endothelial cells have the ability to synthesize nitric oxide.
