局部应用天然与合成冰片的眼刺激性与长期毒性 被引量:17

摘要 目的:观察天然冰片及合成冰片对兔眼刺激性及长期毒性作用。方法:Draize眼刺激性实验。长期毒性实验:32只白色家兔随机分为8组,6个实验组双眼分别滴1,0.5,0.25g/L天然或合成冰片滴眼液,4次/d,连续4wk。空白对照组不用药,溶酶对照组仅用赋形剂。末次点眼后取血做血常规及血生化,计算脏体比系数,取眼球各组织做病理学检查。结果:天然冰片与合成冰片滴眼液对眼局部无刺激性,与对照组相比血常规与血生化检测指标无显著差异,肝脾肾的脏器系数无差异。光镜及电镜结果显示实验组眼球无显著病理改变。结论:兔眼对天然冰片与合成冰片有较好的耐受性,长期应用无眼局部病理变化及肝肾功能损伤。冰片有望做为渗透促进剂广泛用于滴眼液配方。 AIM : To observe the irritation and chronic toxicity effect of natural borneol or synthetic borneol on rabbit eyes. METHODS: Draize eye irritation test was carried out. Chronic toxicity experiment: 32 rabbits were randomly divided into 8 experimental groups. Rabbits in 6 experimental groups received topical 50μ L 1, 0.5g/L, and 0.25g/L natural borneol or synthetic borneol eye drops, respectively, 4 times daily for 4 weeks. Rabbits in control group 1 received nothing as normal control and rabbits in control group 2 received 50 uL of vehicle without borneol as negative control. Hematological parameters and organ coefficient were detected. Histopathological examination was made by light microscope and transmission electron microscopy. RESULTS: Natural borneol and synthetic borneol could not lead to eye irritation. Compared with the control group, blood routine biochemical parameters and organ body coefficient of liver spleen and kidney of experimental group showed no significant differences. Histopathological examination indicated no change in experimental group. CONCLUSION:There is a well tolerance on rabbit eye to natural borneol and synthetic borneol. Borneol has value for incorporation into ocular formulations.
