摘要 目的观察复方保肝宁对HSC-T6细胞凋亡的影响,以进一步探讨保肝宁抗肝纤维化的作用机理。方法引进HSC-T6细胞株并传代培养。用复方保肝宁给正常大鼠灌胃,制备药物血清,温育培养细胞。用吖啶橙染色法和流式细胞仪测定细胞凋亡。结果正常大鼠血清培养组HSC-T6细胞核DNA为黄色或黄绿色均匀荧光,自身凋亡率为(9.67±0.57)%,复方保肝宁药物血清一倍、两倍剂量组细胞核或细胞质内可见致密浓染的黄绿色染色,甚或见黄绿色碎片,凋亡率分别为(22.73±0.78)%,(23.97±2.50)%,明显高于正常对照组,差异有显著性(P<0.01)。结论体外培养的HSC具有一定程度的自身凋亡发生,复方保肝宁的促肝星状细胞凋亡作用可能是其抗肝纤维化作用的主要机理之一。 Objective To investigate the effect of Chinese herb compound Baoganning on apoptosis of HSC - T6 and its mechanism. Methods HSC - T6 were introduced into our laboratory and subcuhured. Normal rats were medicated with compound Baoganning and the serum was collected to prepare the medicated serum, which was incubated with the subcuhured HSC -T6. Then, detected the cell apoptosis rate by using the acridine orange staining and flow cytometer. Results The normal nuclei DNA of HSC - T6, cultured in the normal rat serum, was fluorescent yellow or Kelly, and the apoptosis rate itself of HSC - T6 was (9.67 ± 0.57)%. While the nuclei or cytoplasm, cultured in the medicated serum, was pykno -stained Kelly, even could see the Kelly fragment, and the apoptosis rate were (22.73 ± 0.78 )% and (23.97 ±2.50)%, respectively, which were significantly higher than that of blank - control group ( P 〈 0.01 ). Conclusion Compound Baoganning can significantly accelerate the apoptosis of HSC, which may be the main mechanism of Baoganning on anti - hepatic fibrosis actions.
