摘要
目的观察32^P-玻璃微球(32^P-GMS)间质给药后体内生物学分布与代谢。方法120只小鼠分别经肝脏或肌肉注射32^P-GMS,每只(1.80±0.05)MBq/50μl,不同时间点处死取血及脏器计算每克组织百分注射剂量率(%ID/g);12只大鼠肝脏、肌肉每只注射(18.0±0.5)MBq/0.5m1,收集排泄物测累积排泄率。结果小鼠肝脏组给药肝叶%ID/g0时最高为1.38,15min降至0.71,非给药肝叶峰值15min为0.52,4h之后左右肝叶无区别;肺组织计数率值1h内上升,肝肺累积分流率为37.9%。肌肉组注射点%ID/g0时为31.47,15min~8h为25.06—11.92(中值20.97),12h为8.70,24h~14d为3.54—2.02(中值2.51),其他主要脏器放射性接近本底水平。大鼠肝脏组粪便和尿液14d累积排泄率分别为0.0751%和0.0586%;肌肉组分别为0.0401%和0.0385%。结论32^P—GMS间质注射适用于除肝脏以外的体内组织脏器恶性实体瘤的治疗。
Objective To observe the biodistribution and the metabolism of phosphorus-32 glass mierospheres (^32P-GMS) through interstitial administration. Methods Thirty-two P-GMS were injected into liver and muscle of 120 mice respectively by means of interstitial administration with dose of ( 1.80 ± 0.05 ) MBq/50μl. All of the mice were killed at different time points, blood was taken out and major organs were dissected,and the injection dosage rate (% ID/g) was measured. The stool and urine were collected after interstitial injection both in liver and muscle of 12 rats with the dose of (18.0 ± 0.5 ) MBq/ 0.5 ml,then the accumulative radioactivity counts percentage was measured. Results In liver injection group,the highest point of % ID/g was 1.38 at start time after administration, and decreased to 0.71 at 15 min. The % ID/g value at 15 min was 0.52 in none injected liver lobes ,and there was the similar tendency between left and right liver lobes after 4 h. The radioactivity in lung was increased obviously in 1 h, and the liver-lung accumulate split stream rate was 37.9%. In muscle injection group, the highest % ID/g at start time point was 31.47 after administration,25.06 to 11.92 in 15 min to 8 h ( midvalue was 20.97) ,8.70 at 12 h,and 3.54 to 2.02 in 24 h to 14 days (midvalue was 2.51 ). The radioactivity of other organs and tissues was close to the background level. The accumulative radioactivity counts percentage of stool and urine was 0.0751% and 0. 0586 % respectively in rat liver group, and 0. 0401% and 0. 0385 % respectively in rat muscle group. Conclusion ^32P-GMS interstitial administration may have great potential to treat the malignancy solid tumor in all organs and tissues except liver.
