托吡酯对戊四氮点燃慢性癫痫大鼠海马Fas表达的影响 被引量:1

摘要 目的探讨托吡酯(TPM)对戊四氮(PTZ)慢性点燃癫痫大鼠海马Fas表达的影响及其对海马神经元的保护作用。方法48只大鼠随机分为健康对照组(A组)、PTZ单纯致痫组(B组)、TPM40mg/kg干预组(C组)、TPM80mg/kg干预组(D组),通过腹腔注射PTZ建立慢性癫痫模型,观察大鼠行为、脑电图及海马组织学改变,并用免疫组化染色观察Fas蛋白的表达。结果TPM可以明显抑制大鼠的痫性放电,减少其痫性发作级别,C组、D组与B组比较重型发作率明显降低(P<0.01);此外,TPM干预组凋亡相关基因Fas表达明显减低(P<0.01)。结论在PTZ慢性点燃癫痫大鼠模型中,TPM可降低凋亡相关基因Fas表达,从而减轻海马神经元的凋亡程度。 Objective To explore the effect of TPM on Fas expression in hippocampus in pentylenetetrazole (PTZ) induced chronic kindling seizure rats and the protection on neuron of hippocampus. Methods 48 rats were divided into normal control group(group A),PTZ kindling epilepsy group (group B),TPM 40 mg/kg group (group C) anti TPM 80 mg/kg group (group D). Use chronic model of epilepsy induced by FIE intraperitoneal injection. Changes of behavior,EEG recording and hippocampal histology were observed, and Fas expression was determined with the immunohistochemistry method. Results Compared with group B, the seizure activity and its degree decreased significantly( P 〈 0.01) .The expression of Fas and apoptosis significantly increased in group B and TPM could reduce the expression of Fas and apoptosis( P 〈 0.01). Conclusion In PTZ induced chronic kindling rats, TPM could down-regulate the Fas expression in hippocampus,thus decreasing neuron apoptosis.
