器官移植受者外周血中人巨细胞病毒pp67-mRNA的RT-PCR检测 被引量:1

摘要 目的:探讨使用RT-PCR的方法对人巨细胞病毒pp67-mRNA检测的可行性。方法:对38例肾移植受者的外周血样本共68份分别进行免疫荧光技术检测pp65抗原和RT-PCR技术检测人巨细胞病毒pp67-mRNA。结果:在7例受者外周血中的13份外周血样本中检出pp65抗原阳性,并有1例诊断为HCMV病;在3例受者外周血中的3份外周血样本中检出pp67-mRNA阳性,其中包括2例受者的2份样本pp65抗原同时检出为阳性并有1例诊断为CMV病。结论:pp65抗原和pp67-mRNA检测结果不具有同一性,不能使用pp67-mRNA检测取代pp65抗原的检测。使用RT-PCR可以对人巨细胞病毒pp67-mRNA作出定性的判断。 Objective :To investigate detecting feasibilitiy of human cytomegalovirus pp67-mRNA by Reverse transcription-PCR. Methods :The HCMV pp65 antigen from 68 peripheral blood samples collected from 38 kidney transplantation recipients had been tested via Immunofluorescence assay and RT-PCR. Results ..The positive result of HCMV pp65 antigen from 13 samples from 7 kidney transplantation recipients had been tested via Immunofluorescence assay and 1 recipient was clinically diagnosed with HCMV desease. The positive result of pp67-rnRNA from 3 samples from 3 kidney transplantation recipients had been tested via RT-PCR and 2 samples from 2 kidney transplantation recipients were positive by Immunofluorescence assay at the same time. In the 3 kidney transplantation recipients, 1 recipient was clinically diagnosed with HCMV desease. Conclusion:There are different results by Immunofluorescence assay detecting HCMV pp65 antigen and RT-PCR detecting pp67-mRNA. So the detection of HCMV pp65 antigen can't be replaced by thedetection of pp67 mRNA. RT-PCR only can be used for the judgment on pp67-mRNA character.
