强磁重力环境对成骨细胞增殖、形态及其主要元素含量的影响 被引量:8

摘要 目的:研究强磁重力环境(HMGE)对成骨细胞增殖、形态以及主要元素相对百分含量的影响.方法:采用大梯度超导磁体模拟空间重力环境,以细胞计数法和扫描电子显微镜结合能谱仪(SEM-EDS)分别检测强磁失重(0g),强磁常重力(1g)和强磁超重力(2g)三种HMGE对人骨肉瘤细胞(MG-63)及小鼠成骨样细胞(MC3T3)增殖能力、形态以及细胞中钙、钠、镁等元素百分含量的影响.结果:MG-63和MC3T3细胞在0,1,2g和对照环境中培养24h后,MC3T3细胞平均增殖倍数分别为:-(0.21±0.06),0.91±0.17,0.49±0.04和1.08±0.18;MG-63细胞平均增殖倍数分别为:1.03±0.16,0.75±0.06,0.67±0.15和0.44±0.07.与对照组相比较,0,1,2g环境处理组细胞形态发生明显变化,其中0g环境处理组变化最为明显.各环境处理组MC3T3细胞中钙、钠、镁元素百分含量与对照组相比差异不明显.MG-63细胞中钙元素百分含量0,1,2g环境处理组及对照组分别为(9.93±0.98)%,(3.61±1.53)%,(5.30±1.47)%和(7.35±1.92)%,与对照组相比,0g环境处理组MG-63细胞中钙元素含量明显增加(P<0.01),而1,2g环境处理组MG-63细胞中钙元素含量明显下降(P<0.05).各组MG-63细胞中钠元素含量与对照组相比较差异不明显.MG-63细胞中镁元素百分含量0,1,2g环境处理组及对照组分别为:0%,(1.90±0.77)%,(2.50±0.74)%和(2.83±0.21)%.0,1g环境处理组与对照组相比明显下降(P<0.05).结论:HMGE可影响成骨细胞的增殖、形态以及细胞中钠、镁、钙等元素的相对百分含量. AIM: To investigate the effects of high magneto-grav- itational environment (HMGE) on the proliferation, morphology and the contents of main elements in osteoblasts. METHODS : A specially designed large gradient high magnetic field which could produce three apparent gravity levels (0,1 and 2 g) was used to simulate space gravity environment. The effects of HMGE on cell proliferation, morphology and the contents of main elements in os- teoblasts were detected by cell counting assays, scanning electron microscopy and energy dispersive spectrometer after exposure to HMGE for 24 h. RESULTS : After being exposed to 0, 1, 2 g and control environment for 24 h, the average multiple of MC3T3 cell proliferation was- (0.21±0.06), 0.91 ±0. 17, 0.49 ± 0.04 and 1.08 ±0. 18, respectively. The average multiple of MG-63 cell proliferation was 1.03 ±0. 16, 0.75 ±0.06, 0.67± 0.15 and 0.44± 0.07, respectively. Compared to control environment, the morphology of MC3T3 and MG-63 cells exposed to 0, 1 and 2 g environment was dramatically altered. HMGE (0, 1 and 2 g) had no significant effects on the contents of sodium, magnesium and calcium in MC3T3 ceils. The average percentage of calcium in MG-63 ceils under 0, 1, 2 g and control environment was(9. 93 ±0. 98)%, (3. 61 ± 1. 53)%, (5. 30 ± 1.47)% and (7.35± 1. 92)%, respectively. However, the differences of the average percentage of sodium in MG-63 cells between 0, 1,2 g and control groups were not significant. The average percentage of magnesium in MG-63 cells under 0, 1,2 g and control environment was 0%, (1. 90±0. 77)%, (2. 50±0.74) % and (2.83 ± 0.21 ) %, respectively. CONCLUSION: HMGE affects osteoblast proliferation, morphology and the relative contents of sodium, magnesium and calcium.
