异氟醚对缺氧复氧损伤星形胶质细胞的影响

摘要 目的研究异氟醚对缺氧复氧造成的星形胶质细胞损伤的影响。方法培养新生鼠脑皮质星形胶质细胞,缺氧8 h后复氧8 h,然后用异氟醚(0.28、1.4、2.8 mmo/L)处理细胞,用MTT法检测细胞活性和免疫组化方法研究胶质纤维酸性蛋白(GFAP)表达的变化。结果①1.4 mmol/L异氟醚处理的细胞活性比缺氧复氧组高,有显著性差异(P<0.05);②免疫组化结果显示异氟醚处理组GFAP表达量明显高于缺氧复氧组,组间比较差异有显著性(P<0.05)。结论适当浓度的异氟醚对缺氧复氧造成的星形胶质细胞损伤有一定的缓解作用。 Objective To investigate the effect of isofluran on the injury of astrocytes during hypoxia/reoxygenation. Methods Astrocytes were dissociated from cerebral cortex of 0 -2 day -old rats and divided into three groups: normal group, isofluran group and hypoxia/reoxygenation (H8/R8) group. The astrocytes were treated with isofluran at doses of 0.28, 1.4 and 2.8 mmol/L, respectively. MIT was employed to determine the cellular activity and variations of glial fibrillary acidic protein (GFAP) expressions were observed by immunohistochemistry method. Results (1)The cellular activity of the astrocytes treated with 1.4 mmo/L isofluane was higher than that in H8/R8 group. The difference was statistically significant (P 〈 0.05 ). (2)The expression of GFAP in isofluran group was higher than in H/R groups. The differences between the groups were statistically significant ( P 〈 0.05 ). Conclusion Isofluane at suitable concentrations has effect on recovering astroeytes from injury during hypoxia/reoxygenation.
