紫杉醇联合肿瘤坏死因子α诱导粘液表皮样癌细胞凋亡的实验研究

摘要 目的探讨人肿瘤坏死因子α(TNF-α)联合紫杉醇对人黏液表皮样癌细胞系(MEC-1)的抑制增殖作用及与细胞凋亡的关系,为化疗与生物治疗联合应用治疗粘液表皮样癌提供实验依据。方法将人黏液表皮样癌细胞系(MEC-1)和对照组原代培养鼠肺成纤维细胞(LFB)分为9组,每组给予不用药物浓度作用72h。3H-TDR法检测细胞增殖抑制率;流式细胞术检测细胞凋亡率及细胞周期改变;透射电镜观察凋亡细胞形态变化。结果联合用药组的细胞增殖抑制率显著高于单独用药组(P<0.05)。LFB细胞各组的细胞增殖抑制率显著低于MEC-1细胞(P<0.05)。结论TNF-α联合紫杉醇对MEC-1细胞增殖的抑制作用以及诱导凋亡具有协同作用。 Objective To study the effects of combined use of recombinant human tumor necrosis factor α(rhTNF- α) and paclitaxel on the apoptosis of salivary mucoepidermoid carcinoma cells ( MEC-1 ). Methods The MEC-1 cells and primary culture of mouse lung fibroblasts (LFB) were divided into nine groups, each group receiving treatment of different concentrations of (rhTNF-α) and paclitaxel for 72 h. The inhibition ratio of cell proliferation was detected with 3H-TDR. The percentage of apoptotic cells and cell cycle distribution were examined by flow cytometer. The cell morphological change of apoptosis was observed. Results The inhibition ratio of cell proliferation in group of combined use of two drugs was significantly higher than that in either single drug group ( P 〈 0. 05 ). The inhibition ratio of cell proliferation in LFB cell group was significantly lower than that in MEC-1 cell group ( P 〈 0. 05 ). Conclusion TNF-α associated with paclitaxel was synergetic in inhibiting MEC-1 cells and inducing apoptosis.
