目的:研究米非司酮对人早期妊娠绒毛滋养细胞增殖和凋亡的影响。方法:测定正常妊娠吸宫流产和米非司酮流产绒毛滋养细胞增殖和凋亡情况。PCNA免疫组化着色情况指示细胞增殖,DNA缺口原位末端标记检测细胞凋亡情况。结果:正常早期妊娠绒毛细胞滋养细胞增殖指数为268.7±6.7/500,且见细胞浆阳性现象,合体滋养细胞末见PCNA阳性着色;合体滋养细胞和细胞滋养细胞的凋亡指数分别为12.6±4.3/500和2.6±1.3/500;米非司酮作用后,细胞滋养细胞增殖指数降为254.3±7.2/500,细胞浆阳性现象消失,细胞凋亡指数显著增加,合体滋养细胞和细胞滋养细胞分别达到100.6±8.7/500和110.8±11.3/500。结论:米非司酮终止早期妊娠可能是通过抑制细胞增殖和/或促进细胞凋亡来实现的。
Objective: To study effects of mifepristone on proliferation and apoptosis in early pregnant chorionic villi. Methods: proliferation and apoptosis in early pregnant chorionic villi from artificial and mifepristone induced abortion were studied. Marker for proliferation is proliferating cell nuclear antigen (PCNA) immunohistochemical staining, apoptotic cell death was detected using DNA in situ terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick ending(TUNEL). Results: In early pregnant chorionic villi, proliferating index of cytotrophoblasts was 268.7±7.6/500, not only nucleus but cytoplasma were PCNA positive staining, syncytiotrophoblasts was negative. There existed apoptotic cell death in trophoblasts, the ratio of TUNEL positive cell were 12.6±4.3/500 in syncytotrophoblasts and 2.6±1.3/500 un cytotrophoblasts, respectively. After 2 days mifepristone administered, the number of PCNA positive nucleus staining was decreased slightly and PCNA positive cytoplasmic expression was disappeared.In mifepristone group, apoptotic cells was increased significantly, there were110.8±11.3/500 in syncytiotrophoblasts and 100.6±8.9/500 in cytotrophoblasts, respectively. Conclusion: Mifepristone may inhibit proliferation and promote apoptosis of trophoblasts in early pregnant chorionic villi.
