摘要
背景 5脂氧合酶(5-LO)催化花生四烯酸产生白三烯类炎症介质,与高血压、心肌梗死、脑卒中等多种动脉硬化性疾病关系密切。目的探讨5-LO 在高血压大鼠脑动脉的表达以及是否参与原发性高血压的发生、发展过程。方法用免疫荧光、免疫印迹等方法检测5-LO 在自发性高血压大鼠(SHR)脑动脉血管的表达,并与血压正常的 WKY 大鼠对比,观察5、16、28周龄的 SHR 脑动脉5-LO 表达的差异。结果免疫荧光显示5-LO 在 SHR脑动脉平滑肌细胞中表达,免疫印迹结果也进一步确定其在主动脉、脑动脉血管中的表达;5、1 6、28周 SHR 大鼠脑动脉5-LO 表达量与同周龄 WKY 大鼠相比,分别增加2%(P>0.05)、20%(P<0.01)、39%(P<0.01);5-LO 在SHR 大鼠脑动脉的表达量与大鼠周龄俱增,以5周时 SHR 为100%,16周为115%(P<0.05),28周为141%(P<0.05)。结论 5-LO 在 SHR 高血压大鼠脑动脉平滑肌细胞中的表达,可能参与了高血压病脑血管损害的过程。
Background 5-1ipoxygenase(5-LO) catalyzes arachidonic acid to leukotrienes which involved in complicated interactions between atherosclerosis and invading inflammatory cells. Objective To investigate 5-LO expression in cerebral artery and test the speculation that 5-LO is associated with BP elevation in SHR. Methods The 5-LO expression distribution was detected by immunofluorescence and Western blot, and the level of 5-LO expression was compared in spontaneous hypertensive rat(SHR) and Wistar-kyoto(WKY) rat at 5 weeks, 16 weeks and 28 weeks old. Results 5-LO was expressed in rat cerebral smooth muscle cells, and mainly located in cytoplasm. In 5,16, 28 weeks when the BP was elevated in SHR, the levels of 5-LO were increased by 2% (P〉 0.05), 20% (P〈0.05) and 39% (P〈0.01) respectively, compared with the matched age WKY rats. Using 5-LO at 5th weeks of age as reference, the 5-LO expression in cerebral arteries at 16th weeks were increased to 115% (P〈0.05), at 28th weeks to 141% (P〈0.05). Conclusion Our data shows 5-LO was expressed in smooth muscle cell of cerebral artery, and was progressively increased with the age of SHR. The expression of 5-LO in the cerebral artery of SHR is greater than that in WKY.
