电针对脑梗塞大鼠皮质缺血半影区内TGF-β1免疫阳性细胞的形态学影响 被引量:2

摘要 目的观察电针对脑梗塞大鼠皮质缺血半影区内转化生长因子免疫阳性细胞形态学的影响,为探讨胶质细胞参与针刺治病机理提供实验依据。方法线栓法建立大鼠脑梗塞模型,用免疫细胞化学方法显示针刺对大鼠皮质缺血半影区内转化生长因子阳性细胞的形态、分布及数量与非针刺组、假手术组和对照组的比较,寻找电针对TGF-β1阳性细胞的形态学影响。结果①大鼠脑皮质内转化生长因子阳性细胞的形态学观察对照组大鼠脑额叶皮质内可见散在圆形和椭圆形的转化生长因子阳性细胞。呈棕色TGF-β1免疫反应产物沉积在细胞膜。假手术组大鼠额叶皮质内TGF-β1免疫反应阳性细胞的分布和形态与对照组的未见明显组间差异。非电针组大鼠缺血半影区内可见大量肿胀、免疫反应增强的TGF-β1免疫阳性神经元。但其分布与对照组的比未见明显差异。电针组大鼠缺血半影区内TGF-β1免疫阳性细胞的肿胀程度比非电针组的明显减轻,细胞体积变小。在免疫反应程度上比非电针组得明显增强,免疫产物除沉积在细胞膜外,细胞质内亦可见。②大鼠皮质区内转化生长因子阳性细胞的数量统计对照组大鼠额叶皮质TGF-β1阳性细胞数量与假手术组的差异无统计学意义。非电针组的比对照组的明显增加,差异有统计学意义(P<0.01)。电针组大鼠缺血半影区内TGF-β1阳性神经元的数量比非电针组的明显增加,差异有统计学意义(P<0.01)。结论1~2mA强度的电刺激"曲池"和"足三里"穴位能上调大鼠额叶皮质缺血半影区内TGF-β1的表达,上调的TGF-β1可能参与对缺血性神经元损伤的保护过程。 Objective To observe morphological effect of TGF-β1 immonoactive positive cells in penambra area by electroacupuncture stimulation in ischemia rat. Methods The cerebral infarct model of rat was established by thread embolism. Immunocytochemistry method are used to observate the cellular morphological difference and number of TGF-β1 immunoactive positive cells at penambra region of cerebral cortex in rats. Results (1) The morphological observation of TGF-β1 immunopositive cells: In control group TGF-β1 immunopositive cells were round and elliptic in shape and distributed scatteredly in all layers of frontal cortex.. The product of TGF-β1 immunoaction deposited cytomemmbran. There was no obvious difference in morphology and distribution of TGF-β1 immunopositive cells between control and sham operated group. In nonelectroacupuncture group TGF-β1 immunopositive cells were obviously swelling and larger in size than that in control group and appeared weiying ceils as well as pyknosis cells. There was no difference in distribution of TGF-β1 positive cells between nonelectroacupuncture and control groups. In electroacnpuncture group TGF-β1 immunoapositive cells were obviously decreased in the swelling and pyknosis cells than that in nonelectroacupunctnre group and immunoactive were obviously enhanced and the immunoproduct were not only deposite on the cyromemmbrane but also in the cytoplasm. (2) The number of TGF-β1 immunopositive cells There was no differences in number of TGF-β1 immunopositive cells between control group and sham operated group but in nonelectroacupuncture group there were more than that in control group and in electroacupuncture group the number of TGF-β1 immunopositive cells were more than in nonelectroacupuncture grou and the difference was significantly in statistics. Conclusion 1-2mA electroacupuncture stimulation of quchi and zu shan li acupoint can up-regulated NGF-β1 expression at penambra region of ischemic cerebral cortex and up-regulated TGF-β1 may provide protection
