摘要
目的通过长期毒性研究来评价乳化异氟烷的溶血性。方法健康SD大鼠100只,雌雄各半,体重100~160g,6周龄,随机分为5组(n=20):对照组经尾静脉注射生理盐水3ml/kg;溶媒组静脉注射30%脂肪乳3ml/kg;乳化异氟烷高、中、低剂量组分别静脉注射乳化异氟烷132、108和84mg/kg;健康Beagle犬18只,雌雄各半,体重7.4~8.8kg,7月龄,随机分为3组(n=6):对照组经前臂头静脉注射生理盐水2ml/kg;溶媒组静脉注射30%脂肪乳2ml/kg;乳化异氟烷组静脉注射乳化异氟烷135mg/kg。所有动物每天给药1次,连续30d(给药期)。给药期结束时各组随机取半数动物,计数全血红细胞和网织红细胞,测定血红蛋白和总胆红素浓度,观察脾和肾组织病理学结果,Beagle犬行骨髓细胞学检查,各组剩余动物继续观察2周后(恢复期结束后)重复上述实验。结果与对照组比较,乳化异氟烷高剂量组SD大鼠恢复期结束后全血红细胞计数降低(P〈0.05),其余各指标差异无统计学意义(P〉0.05);各组Beagle犬给药期和恢复期结束后各指标差异均无统计学意义(P〉0.05);SD大鼠和Beagle犬脾和肾组织病理学结果未见异常,Beagle犬骨髓细胞学检查未见异常。结论长期毒性研究时乳化异氧烷未引起大鼠和犬溶血反应。
Objective To determine whether emulsified isoflurane causes hemolysis during long-term toricity study. Methods Test Ⅰ : one hundred 6-month old SD rats of both sexes weighing 100-160 g were randomly divided into 5 groups ( n = 20 each) : Ⅰ control group received normal saline (NS) 3 ml/kg iv daily for 30 consecutive days; Ⅱ vehicle group received 30 % fat emulsion 3 ml· kg^-1·d^-1 iv × 30 d; Ⅲ , Ⅳ, Ⅴ high, medium, low dose emulsified isoflurane groups received emulsified isoflurane 132, 108 and 84 mg/kg iv × 30 d respectively. Test Ⅱ :eighteen 7-month old beagle dogs were randomly divided into 3 groups (n = 6 each): 1 control group received NS 2 ml·kg^-1·d^-1 iv × 30 d; Ⅱ vehicle group received 30 % fat emulsion 2 ml·kg^-1·d^-1 iv × 30 d and Ⅲ emulsified isoflurane group received emulsified isoflurane 135 ml·kg^-1·d^-1 iv ×30 d. At the end of the last iv injection and 2 weeks after the last injection, blood samples were collected from abdominal aorta for determination of RBC and reticulocyte counts and Hb and total bilirubin concentrations in half of the animals in each group. The animals were then killed and spleen and kidney specimens were obtained for histopathologic examination. Results Except slight reduction in RBC count in high-dose emulsified isoflurane group there were no significant differences between emulsified isoflurane and control groups. Histopathologic examination did not find any specific lesions in the spleen and kidney. Conclusion Emulsified isoflurane does not cause hemolysis in rats and dogs during long-term toxicity study.
