摘要 目的:探讨促血管生成素-1(angiopoietin-1,Ang-1)在5/6肾切除大鼠肾脏中的表达变化及其与肾脏微血管病变的关系。方法:复制5/6肾切除大鼠模型,设假手术组和5/6肾切除模型组,在1、2、4、8和12周,采用免疫组化观察肾脏Ang-1和CD31表达变化以及RT-PCR观察肾脏Ang-1 mRNA表达。结果:模型组2、4和8周时肾脏Ang-1 mRNA表达显著上调,12周时低于假手术组;模型组2周~8周免疫组化显示肾小球Ang-1阳性着染均显著高于假手术组,峰值在4周~8周,12周后逐渐下调;模型组2周~12周肾小球CD31表达逐渐减少。结论:5/6肾切除大鼠肾脏存在Ang-1和CD31表达的改变,此改变参与了残肾微血管结构的变化。 Objective-To investigate the expression of angiopoietin- 1 (Ang- 1 ) in 5/6 subtotal nephrectomy mrs model, and explore that those changes are whether or not correlated to the renal microvasculature injury. Methods: Rat model were estab- lished in adult male 813 rats by 5/6 nephrectomy. The renal function and histopathological changes were evaluated at 1,2,4,8 and 12 weeks after operation finished. The expressions of Ang - 1 and CD31 in renal were detected by immunohistochernistry. Ang - 1 mR- NA was detected by RT- PCR at the same time- points. The levels of Ang- 1 and CD31 were quantified by computer image analysis. Resultss:Ang- 1 mRNA on renal tissue in rats with STN was significantly up regulated at 2,4 and 8 weeks compared with that in STN group at other time points or in CTL group at the same time - points, while decreased evidently at 12 week compared with that in CTL group. From 2 - week to 8 - week,Ang - 1 was outstandingly expressed on glomeruli in rats with STN, the number of Ang - 1 staining in STN glomeruli increased significantly compared with that in CTL group, being maximal at 4 and 8 weeks, and subsequently decreased after 12 week. Conclusion: The expression of Ang- 1 on renal tissue in rats with STN is exchanged, which are early up regulation and later down regulation. Those changes are correlated with renal microvasculature injury.
