摘要
目的:探讨厄多司坦是否对后肢缺血再灌注所致的肺损伤具有保护作用。方法:将24只成年雄性大鼠随机分为3组:对照组、缺血再灌注组及厄多司坦组。手术后检测肺组织一氧化氮(NO)含量,髓过氧化物酶(MPO)、腺苷脱氨酶(ADA)以及黄嘌呤氧化酶(XO)的活性。同时检测支气管肺泡灌洗液NO含量、MPO活性。结果:与对照组相比,缺血再灌注组肺组织中ADA及XO活性增加(P<0.05)。缺血再灌注组NO水平高于对照组(P<0.05),而厄多司坦组NO水平无改变。与对照组及厄多司坦组相比,缺血再灌注组MPO活性增加(P<0.05)。与对照组相比,缺血再灌注组支气管肺泡灌洗液MPO活性升高(P<0.05)。厄多司坦组支气管肺泡灌洗液MPO活性显著低于缺血再灌注组(P<0.05)。与对照组相比,缺血再灌注组及厄多司坦组支气管肺泡灌洗液NO水平升高(P<0.05)。不过,相对于缺血再灌注组,厄多司坦治疗组支气管肺泡灌洗液NO水平显著降低。结论:厄多司坦可降低大鼠后肢缺血再灌注后氧化应激性损伤及中性粒细胞堆积,从而对肺损伤有一定的保护作用。
Objectlve:The goal of this experimental study was to investigate whether erdosteine had a protective effect against lung injury as a remote organ after hind-limb ischemia-repeffusion (I/R). Methods:The rats were divided into three groups:control, I/R, and I/ R erdosteine groups. After the experimental procedure,nitricoxide(NO) levels,Myeloperoxidase(MPO),adenosine deamlnase(ADA),and the activities of xanthine oxidase (XO) were determined on the lung tissue. The levels of NO and activities of MPO were also measured on the bronchial alveolar lavage (BAL). Results:The lung tissue ADA and XO activities were increased in the FR group compared with those in the control group (P〈0.05). In the FR group,the level of NO was higher than that in the control group (P〈0.05), whereas the erdosteine treatment did not alter the NO level(P〈0.05).The MPO activity increased after FR in the FR group compared to that in both control and I/ R erdosteine groups(P〈0.05). The activity of MPO increased in the IR group in comparison with that in the control group in BAL(P〈0.05). The activity of MPO in the FR erdosteine group was significantly lower than that in the I/R group in BAL (P〈0.05). NO level increased in all I/R groups compared to that in control group in BAL (P〈0.05). However, treatment of erdosteine significantly decreased NO level compared to FR group(P〈0.05). Conclusion:As a result, erdosteine may be a protective effect for lung injury,decreasing oxidative stress and neutrophil accumulation after hind-llmb I/R in rats.
