摘要
【目的】探讨生长抑素区域动脉灌注对重症急性胰腺炎(SAP)大鼠胰腺腺泡细胞凋亡的影响。【方法】SD大鼠64只,随机分为假手术组(A组),SAP组(B组),生长抑素外周静脉注射组(C组),生长抑素区域动脉灌注组(D组)。测定各组大鼠的血清淀粉酶(Amy),观察胰腺病理变化,并检测胰腺腺泡细胞的凋亡率。【结果】①制成SAP模型后,各组Amy水平显著上升,而C组和D组可使Amy下降,以D组下降更为明显。②C组和D组生长可使胰腺腺泡细胞凋亡率明显上升,胰腺病理损害程度减轻,以D组作用更强。【结论】①生长抑素可诱导SAP大鼠胰腺细胞凋亡,减轻胰腺病理损害,是治疗SAP的有效方法。②生长抑素区域动脉灌注是治疗SAP的增效途径。
[Objective]To study the effects of regional intra-arterial infusion with somatostatin on apoptosis in rats with severe acute pancreatitis (SAP).[Methods]SD rats were divided into four groups randomly: Sham-operative group (group A), SAP group (group B), group of peripheral vessel infusion of somatostatin (group C), group of regional intra-arterial perfusion of somatostatin (group D). The contents of serum amylase and pancreatic histologic changes were observed and apoptosis of pancreatic aeinar cells was detected by TUNEL. [Results]①Serum amylase level of each group after establishment of model was much higher than that of before it. Treatment with somatostatin decreased serum amylase level. Regional intra-arterial perfusion of somatostatin had the most remarkable effectiveness. ②After treatment with somatostatin, the apoptotic ratio of acinar cells was increased and the pathological damage severity of pancreas was significantly attenuated, especially in group D. [Conclusion]①Treatment with somatostain eould improve the apoptotic ratio of acinar ceils in SAP rats and has beneficial effects on pancreas damage. ②Regional intra-arterial perfusion is one of the means of improving therapeutic effect of drugs against SAP.
