摘要
【目的】研究通过视黄酸(RA)对大鼠急性放射性肺损伤的干预作用,阐明其对放射性肺损伤肺组织表皮生长因子受体(EGFR)表达的影响。【方法】健康雄性Wistar大鼠80只随机分为4组:正常对照组(A组)、单纯给药组(B组)、单纯照射组(C组)和照射加给药组(D组),每组20只。C、D两组大鼠行6MV-X线15Gy全胸野照射,B、D两组大鼠给予20mg/(kg·d)RA制剂灌服,于照射后第1、2、4、8周取右肺组织行HE和Masson染色,免疫组化方法检测EGFR的表达。【结果】HE和Masson染色提示照射后的1周始肺泡腔有炎性细胞渗出,继之间质水肿,4及8周出现肺泡腔变小甚至结构破坏,局部实变,肺间质出现胶原纤维;B组与A组比各时间段的病理变化无明显差别,但D组与C组比较大鼠肺炎和肺水肿减轻,肺组织胶原纤维量减少。EGFR免疫组化染色显示C组与A组相比在各时间段EGFR表达均明显增强(P<0.001),D组与C组比第4、8周EGFR表达明显减弱(P<0.001)。【结论】EGFR参与放射性肺损伤的发生、发展,而RA从蛋白质水平有效抑制EGFR表达,为治疗放射性肺损伤提供了实验依据。
[Objective] To investigate the intervention effect of retinoic acid (RA) on acute radiation-induced lung injury ,then to illuminate that RA effects the expression of epidermal growth factor reeeptor(EG-FR) in rat models . [Methods] Eighty Wistar rats were randomly divided into four groups with 20 rats in each: control group( group A),RA treatment group(group B),radiation group(group C) and radiation combined with RA treatment group(group D). Rats in group C and D were radiated with 6MV-X ray linear accelerator at a dose of 15Gy thoracically, and rats in group B and D were given RA by gastrogavage at a dose of 20mg/( kg·d), and rats in group A and C were given physiological saline at the same dose until they were killed. In 1, 2, 4 and 8 weeks, the pathological change of lung tissues and the expression of EGFR were detected with HE, Masson and immunohistochemistry staining, respectively. [Results] HE and Masson staining showed that only one week after radiation, the alveolar spaces had exudative inflammatory cells, following interstitial edema, and at 4 and 8 weeks, alveolar space shrank, even its structure was destroyed with local consol, and pulmonary interstitium appeared collagen fibers. There was no significant difference in pathological change be tween group A and B, but pneumonia and pulmonary edema in group D were obviously less than those in group C, and the expression of EGFR was significantly higher in group A and C at all times(P〈0.001) compared with control group, while it was lower in group D compared with group C at 4 and 8 week (P〈0. 001). [Conclusion] EGFR may participate in the process of radiation induced lung injury, and RA could effectively inhibit the expression of EGFR in protein level.
