摘要
离心脱脂后的牛乳,采用离子交换层析、疏水层析、透析等生物化学技术进行分离纯化,然后对分离纯化得到的产物进行SDS-PAGE电泳、免疫印迹技术初步鉴定,得到相对分子量约为60kDa、具有活性的骨桥蛋白。主要阐述牛乳中骨桥蛋白的分离纯化及初步鉴定的方法,为骨桥蛋白功能性质的研究奠定了基础。
The centrifugal defatted bovine milk was separated and purified by biochemical technique, including ion -exchange chromatography, hydrophobic chromatography and dialysis. The fractions containing osteopontin was identified by SDS-PAGE and Western Blot. Osteopontin with high purity was obtained and its molecular weight was 60kDa. This article mainly elaborate the method about isolation, purification and primary characterization of osteopontin from bovine milk, this provides basis for next step research about the functionality of osteopontin.
