摘要
目的:建立HPLC-ELSD法测定山药中尿囊素的含量。方法:以20%乙醇超声提取,用高效液相色谱-蒸发光散射检测器测定,Hypersil C_(18)(200 mm×4.6 mm,5μm)色谱柱,以甲醇-水(10:90)为流动相,流速为0.8 mL·min^(-1),雾化温度为35℃,蒸发温度为50℃,载气流速为1.5 L·min^(-1)。结果:尿囊素在5.5~16.5μg范围内线性关系良好,方程为Y=1.283X+0.3319(r=0.9992),回收率为101.0%(RSD=2.3%)。结论:该方法专一稳定,明显优于现有测定法,可用于山药中尿囊素的测定。
Objective:To study the quantitative method for allantoin in Rhizoma Dioscoreae by HPLC -ELSD. Method:The column used was a packed with 5μm stationary phase Hypersil C18(200 mm ×4.6 mm,5μm). The mobile phase consisted of methanol -water( 10: 90) , with a flow rate of 0.8 mL· min^- 1, the contents of allantoin were measured. At a temperature of drift tube of 50℃ and the nebulizer nitrogen flow rate was 1.5 L·min^ - 1. Resuit:The linear range of the component was 5.5 - 16.5 μg, the regress equation: Y = 1. 283X + 0. 3319 ( r = 0. 9992). The average recovery of allantoin in crude material was 101.0% ( RSD = 2.3% ). Conclusion:The method is reliable, simple and single - minded, and it can be used for the determination of allantoin in Rhizoma Dioscoreae.
