摘要
本实验探讨用BA-ELISA方法检测培养的平滑肌细胞清道夫受体的方法。第10~15代平滑肌细胞接种于96孔板,加入氧化低密度脂蛋白培养一定时间,固定,按BA-ELISA法测定细胞结合及摄入氧化低密度脂蛋白的量,以反映清道夫受体的活性。结果表明,随培养时间延长至6~24h,清道夫受体活性已明显可测。说明第10~15代的平滑肌细胞在一定条件下可表达清道夫受体活性。
The scavenger receptor activity of smooth muscle cells(SMCs) was detected by the method of BA-ELISA- The SMCs in 10~15 generations were seeded in 96-well plates; then oxidized low-density lipoprotein (Ox-LDL) was added to incubate for a certain period of time. Scavenger receptor activity of SMC was shown by Ox-LDL binding and uptake according to BA-ELISA.The results indicated that SMCs in 10~15 generations could express scavenger receptor activityby this method in which Ox-LDL should be incubated with SMCs at least 6~24 h.
