摘要
体外建立并扩繁了早梨18号的无性系,以试管苗叶片为外植体。研究了基本培养基、植物生长调节剂组合、暗培养时间对叶片不定芽诱导与植株再生的影响。结果表明:早梨18号叶片在适宜的条件下可发生大量不定芽;暗培养预处理有利于提高叶片的再生频率,以暗培养15 d为宜;叶片在NN 69+6-BA 5.0 mg/L+NAA 0.5 mg/L和NN 69+TDZ 1.0 mg/L+IBA0.5 mg/L的培养基上分别获得了56%和54%的叶片再生频率。
The asexal line of "Pear cv premature 18" was established and propogated,leaves of in vitro "Pear cv premature 18" were used as explants.Effects of basicmedium,plant growth regulator,dark culture period on adventitious buds induction and plantlet regeneration were studied.Results showed that: In the best condition,many adventitious buds regenerated from leaves.Dark pretreatment could promote leaves regeneration rates,15 days was the optimum period.In themedium of NN 69+6-BA 5.0mg/L+NAA 0.5mg/L and NN 69+TDZ 1.0mg/L+IBA 0.5mg/L,leaf regeneration frequency was 56% and 54% respectively.
