摘要
目的:研究用离子交换纤维分离纯化葛根素的方法和工艺。方法:将葛根的浸提液在预处理过的纤维上吸附过滤,再用70%乙醇洗脱,得纯化的葛根素。结果:最佳浸提葛根素的工艺参数:60%的乙醇浸提液pH=8,浸提温度60℃,提取时间3 h,浸提2次。得到的葛根素粗品收率为10.92%,纯度为4.56%。最佳分离纯化葛根素的工艺参数:在上柱药液pH=9,流速3 BV.h-1,药液浓度为1.703 mg.mL-1,体积为50 mL的条件下,解吸剂为70%乙醇和2 mmol.mL-1乙酸的混合液(体积比4∶1)4 BV,洗脱流速6 BV.h-1。葛根素经离子交换纤维分离提纯后,产品纯度由4.56%提高到23.25%,产品总收率由10.92%降到2.89%。离子交换纤维对葛根素的吸附率为95.52%,解吸率为96.29%。结论:离子交换纤维分离纯化葛根素方法可行。
Objective：To separate and purify puerarin from the root of kudzu vine by ion exchange fiber. Methods ： After the lixivium of root of kudzu vine was adsorbed and filtrated by ion exchange fiber, Ion exchange fiber is washed with 70% ethanol and refining puerarin was obtained. Results：The optimized puerarin extraction process parameters were as follows ：the sample was extracted with 60% ethanol （pH = 8） at 60 ℃ ,the extract was remained for three hours,and the sample was extracted two times. The crude yield of puerarin was 10.92% ,the purity was 4. 56%. The optimized purification process parameters were as follows：the extraction solution efflux velocity was at 3.0 BV.h^-1, the extraction puerariae solution concentration was 1. 703 mg.mL^-1 at pH = 9.0, the sample volume was 50 mL,the desorbing agent consisited of 70% ethanol and 2 mmol.mL^-1 acetic acid（4：1 ） volume 4 BV,the eluting efflux velocity was 6 BV.h^-1. After puerariae was separated and purified,the purity was increased from 4.56% to 23.25% , the yield is decreased from 10.92% to 2.89%. The adsorption rate of ion exchange fiber on puerarin 95.52% , the desorption rate was 96.29%. Conclusion ：Ion exchange fiber can be used to separate and purify puerarin.
