摘要
目的研究幽门螺杆菌(HP)培养滤液对SGC7901胃癌细胞表达hTERT、Bcl-2蛋白的影响。方法在HP培养滤液诱导前后,采用流式细胞仪检测SGC7901胃癌细胞hTERT、Bcl-2蛋白的表达。结果对照组SGC7901胃癌细胞表达hTERT和Bcl-2蛋白的阳性细胞数和荧光指数显著低于经HP培养滤液诱导的SGC7901胃癌细胞组(P<0.05)。经HP培养滤液处理的SGC7901胃癌细胞24、36、48 h表达hTERT和Bcl-2蛋白的阳性细胞数和荧光指数呈递增趋势,且相互间有显著差异(P<0.05)。结论HP感染不仅通过激活端粒酶促进细胞永生化,而且通过促进Bcl-2表达使细胞凋亡减少,进而促进胃癌的发生、发展,这可能是HP感染致癌的重要机制之一。
OBJECTIVE To study the effect of H. pylori incubating filtrate on the expression of hTERT and Bcl - 2 protein in human 7901 gastric carcinoma cells. METHODS Flow cyto metryanalysi (FCM) was used to study the hTERT and Bcl -2 protein expres- sion in SGC790l ceils under the stimulation of low concentration of H. pylori incubating filtrate. RESULTS The number of cells and the fluorescent index of expressing hTERT and Bcl -2 protein in control SGC7901 group were significantly lower than that in SGC7901 cells cultured with H. pylori culturing filtrate group( P 〈 0.05 ). The number of cells and the fluorescent index of expressing hTERT and bcl -2 protein in 5GC7901 cells cultured with H. pylori culturing filtrate group took an increasing tendency at 24,36,48 hours (P 〈 0. 05 ). The differences in different group were significatnt ( P 〈 0.05 ). CONCLUSION H. pylori infection could not only activate telomerase to lead cells to be immortal but also promote the expression of Bcl - 2 protein to decrease the apoptosis of gastric mucous membrane cells, and then promote the genesis and devolepment of gastric cancernoma. It may be a main caneerigenic mechanism of H. pylori.
