摘要
目的建立测定丹参提取液中丹参素、丹参酮ⅡA含量的HPLC法。方法用Hypersil-ODS色谱柱分别进行测定,丹参素以甲醇-醋酸为流动相,检测波长281 nm,丹参酮以甲醇-水为流动相,检测波长268 nm。结果丹参素在0.1-2.0μg范围内线性关系良好(r=0.999,P〈0.01),高、中、低3种浓度日内、日间精密度小于10%;丹参酮ⅡA在0.25-4.00μg范围内线性关系良好(r=0.999,P〈0.01),高、中、低3种浓度日内、日间精密度小于10%。样品中丹参素的含量:水提取液为1.38 mg/g,体积分数0.50的乙醇提取液为0.48 mg/g,体积分数0.95的乙醇提取液为0.28 mg/g。丹参酮ⅡA的含量:水提取液为0.04 mg/g,体积分数0.50的乙醇提取液为0.64 mg/g,体积分数0.95的乙醇提取液为7.57 mg/g。结论该方法快速、准确、简便,可用于丹参提取液中丹参素和丹参酮ⅡA含量的测定。
Objective To establish HPLC technique for determining the contents of Danshensu and Tanshinone II A in Danshin extract. Methods Hypersil-ODS chromato bar was used for detection. For Danshensu: its mobile phase was CH3OH- CHaCOOH, detection wavelength was 281 nm, Tanshinone Ⅱ A: its mobile phase was CH3OH H2O the detection wavelength was 268 nm. Results The linear ranges of Danshensu was 0. 1-2.0 μg (r=0. 999,P〈0.01) and Tanshione was 0. 25- 4.00 μg (r=0. 999,P〈0.01,) respectively. The RSDS of within day were less than 10%. Conclusion A quick, exact and con- venient method is established to determine the contents of Danshensu and Tanshinone in extract of Tanshin.
