摘要
目的:探讨整合素连接激酶(ILK)和α-平滑肌肌动蛋白(α-SMA)在肾间质纤维化中的作用及意义。方法:建立肾间质纤维化动物模型,随机分为假手术组、模型组和正常对照组,术后1、3、7、14d处死各组大鼠,取梗阻侧肾做HE、PAS及Masson染色,并用免疫组化方法检测ILK和α-SMA的表达,RT-PCR方法检测ILKmRNA表达。结果:ILK主要表达于病变肾小管上皮细胞和一些肾间质细胞胞浆,并且随间质病变的加重,表达量增加,损害评分增大。在正常组织中,α-SMA仅表达于血管壁,但在损害间质中,表达于损害的肾小管上皮细胞和间质细胞,且随着损害程度的加重而增加。ILK、α-SMA与间质纤维化程度呈正相关,其在间质中的表达量也成正相关。结论:随着间质病变程度的加重,ILK、α-SMA的表达量明显增加,说明其与肾间质纤维化的形成有密切关系。
Objective To explore the expressions and significanees of integrin-linked kinase (ILK) and α-smooth muscle actin(α-SMA) in rat models of renal interstitial fibrosis. Methods The rat models of renal interstitial fibrosis were built and assigned randomly into sham-operated group, model group, and control group, which were sacrificed at day 1,3,7,14 after the surgery, and the tissues of the obstructed kidney were stained by HE, PAS, and Masson. The expressions of ILK and α-SMA in the obstructed kidney was assessed by immunohistochemistry, and the expression of ILK mRNA was detected by RT-PCR. Results ILK was expressed mainly in tubular epithelial cells and interstitial cells, and was increased with the severity of lesions, α-SMA was only expressed in vascular wall in the normal tissue, but was expressed in tubular epithelial cells and interstitial cells and was increased with the severity of lesions in damaged interstitial. There were positive correlations between the expression of ILK, α-SMA and the degree of interstitial fibrosis, and the expressions of them were positively correlated. Conclusion The expressions of ILK and α- SMA are increased with the severity of lesions, suggesting that they are correlated with renal interstitial fibrosis.
