黑素瘤抗原-3基因及蛋白在非小细胞肺癌中的表达

摘要 目的检测黑素瘤抗原-3(MAGE-3)mRNA及其编码的肿瘤抗原在非小细胞肺癌(NSCLC)中的表达,探讨以该抗原作为疫苗对NSCLC患者进行免疫治疗的可能性。方法用RT-PCR方法和流式细胞术(FCM)对3种人肺癌细胞系和56例NSCLC标本及相邻正常肺组织标本MAGE-3 mRNA及蛋白的表达情况进行研究。结果3种人肺癌细胞系均表达MAGE-3mRNA及蛋白;56例NSCLC标本中,30例表达MAGE-3 mRNA,26例表达MAGE-3蛋白,而相邻正常肺组织均不表达MAGE-3mRNA或蛋白。结论MAGE-3 mRNA及蛋白在NSCLC中具有较高的表达频率,提示该抗原有望成为对NSCLC患者进行免疫治疗的适宜靶点。 Objective To determine the expression of melanoma antigen-3 mRNA and its encoding tumor antigen in non-small cell lung cancer(NSCLC) and to explore the possibility of using this antigen as a vaccine for immunotherapy in NSCLC patients. Methods The expression of MAGE-3 mRNA and protein in 3 human lung cancer cell lines and 56 NSCLC samples together with the adjacent normal lung samples was examined by RT-PCR and flow cytometry (FCM). Results All 3 human lung cancer cell lines expressed MAGE-3 mRNA and protein. Of the 56 NSCLC samples, 30 cases expressed MAGE-3 mRNA and 26 cases expressed MAGE-3 protein, but none of the adjacent normal lung samples expressed MAGE-3 mRNA or protein. Conclusion MAGE-3 mRNA and protein are expressed at a high frequency in NSCLC samples, and this antigen may possibly be the suitable target for immunotherapy in NSCLC patients.
