摘要 目的研究化瘀理肺方对肺间质纤维化大鼠肺组织中转化生长因子β(TGF-β)、Smad3、Smad7蛋白表达的影响。方法对照组在气管插管内注入生理盐水,其余大鼠注入博莱霉素建立肺纤维化模型。分别给予大鼠化瘀理肺方药物(中药组)、生理盐水灌胃(模型组、对照组)、氢化可的松琥珀酸钠腹腔注射(激素组)进行干预,并于给药后14、28 d处死,取肺组织行HE染色观察病理变化,免疫组化法测定肺组织TGF-β、Smad3、Smad7蛋白表达情况。结果与模型组和激素组比较,中药组肺泡炎和肺纤维化程度及TGF-β、Smad3蛋白表达均明显降低,Smad7蛋白表达升高。结论TGF-β、Smad3、Smad7在实验性大鼠肺纤维化的形成中起重要作用,化瘀理肺方可通过调节这些蛋白的表达,从而起到防治肺纤维化的作用。 Objective To explore the influesnce of Huayulifei on the expression and significane of TGF-β,Smad3 and Smad7 on bleomycin-induced pulmonary interstitial fibrosis in rats. Methods The rats were randomly divided into control group, model group, Huayulifei group and hydrocortisone group. The rats were then intratracheally dripped with bleomycin in experimental groups ,while 0.9% saline in control group. Treatments were started from the next day with a daily oral Huayulifei in Huayulifei group,intraperitoneal hydrocortisone sodium succinate in hydrocortisone group, and oral saline in control group and model,respectively. Rats were killed at 14 and 28 days after bleomycin, lungs isolated, hematoxylin and eosinstaining,and expression of TGF-β,Smad3,Smad7 protein was measured by immunohistochemieal technique. Results alveohtis and pulmonary fibrosis in Huayulifei group was alleviated signgicantly compared with those in model and hydroeortisone group. The expressions of TGF-β, Smad3 protein were significantly lighter in model group and hydrocortisone group, the expressions of Smad7 protein were significantly higher in model group and hydrocortisone group. Conclusions TGF-β, Smad3, Smad7 can enduce important effect on blemyeln-induced pulmonary fibrosis in rats, Huaytdifeifang can enduce prevention and cure by regulate the expression of these protein.
