摘要
目的探讨帕金森病脑损伤对神经干细胞增殖的影响及银杏叶提取物对其干预作用。方法C57BL/6小鼠随机分成正常组、模型组和银杏叶提取物治疗组。采用1-甲基-4-苯基1,2,3,6四氢吡啶腹腔注射建立帕金森病小鼠模型,利用免疫组化和免疫双标技术观察不同组别小鼠齿状回神经干细胞增殖、分化和迁移规律以及银杏叶提取物对其的干预作用。结果模型组第28天,齿状回增殖性核抗原(PCNA)阳性细胞表达显著增多,而银杏叶提取物治疗组的PCNA阳性细胞比正常组增多,但比模型组少。模型组和治疗组第7、28天小鼠齿状回神经上皮干细胞蛋白(Nestin)阳性细胞均明显增加,其中治疗组第7天增加最明显。模型组和治疗组PCNA/Nestin双标细胞的数量在第7、28天均明显增多,治疗组第28天最明显。结论帕金森病脑损伤本身可诱导脑内神经干细胞的增殖,而银杏叶提取物可进一步促进其增殖并可能促进其迁移。
Objective To explore the effect of brain injuries of Parkinson's disease on proliferation of neural stem cells in brain and role of ginkgo biloba extract. Method C57BL/6 mice were randomly divided into normal group, model group and treatment group . Parkinson's disease model was made by i. p. 1 - methyl ,4 - phenyl - 1 ,2 ,3 ,6 tetrahydropyridine. Proliferation and migration of neural stem cells in dentate gyms in every group was observed by immunohistochemistry and double labeled immunohistochemistry technique. Result On 28th day , the number of the PCNA positive cells was increased significantly in model group(MG) and ginkgo biloba extract group( EGBG), but the increasing in MG was more than that in EGBG. The number of the nestin positive cells in dentate gyms was increased in MG and EGBG on 7^th and 28^th day, and the increasing in EGBG was the most obvious on 7^th day . The number of the PCNA/Nestin double positive ceils in dentate gyms was increased in MG and EGBG on 7^th and 28^th day, and the increasing in EGBG was the most on 28^th day. Conclusion The brain injuries of Parkinson's disease may induce proliferation of neural stem cells in brain . Ginkgo biloba extract could promote proliferation and migration of neural stem ceils in brain of Parkinson's disease model mice.
