摘要
目的观察铅对SD大鼠三叉神经细胞内游离钙的影响及其对电压依赖性钙通道作用,以探讨铅对三叉神经细胞毒性作用的机制。方法用全细胞膜片钳方法检测铅对三叉神经细胞电压依赖性钙通道的作用,并用激光共聚焦和FLUO-3/AM荧光探针标记技术,从单个细胞水平检测铅对胞内游离Ca2+瞬间动态变化的影响。结果铅可致三叉神经细胞内游离钙[Ca2+]i升高,铅又可抑制三叉神经细胞电压依赖性钙通道电流ICa。结论铅致三叉神经细胞内游离钙[Ca2+]i升高作用与胞内钙库释放有关;铅抑制三叉神经细胞电压依赖性钙通道ICa作用与其抑制高电压激活(HVA)钙通道有关。
Objective To investigate the effects of lead on intracellular free calcium and voltage-dependent calcium channel in rat trigeminal nerve cells and to explore the toxicity mechanism of lead in rat trigeminal nerve cells. Methods Whole cell patch clamp technique was used to determine Ica Of voltage-dependent calcium channel in trigeminal nerve cells. Intracellular free calcium was measured to explore [Ca^2+ ]i dynamic changes from a single cell level by laser scanning confocal microscopy and fluorescence probe technique. Results Lead resulted in increase of intracellular free calcium concentration in trigeminal nerve cells. Also, lead inhibited voltage-dependent calcium channel currents ICa in trigeminal nerve cells. Conclusions The effect that the increase of intracellular free calcium concentration in trigeminal nerve cells was induced by lead was related to the release of intracellular calcium stores. And the voltage-dependent calcium channel ICa inhibited by lead was dependent on high-voltage-activated(HVA) calcium channels.
